He was raised by her mother alone and her name is Calsinah Manzini together. Sipho grew up with his sibling Bheki Manzini. His father died when he was very young. Manzini is married to his wife Nhlanhla Manzini. They have been happily married for 15 years and he is a loving husband to her . They have two children and both namely Nhlosenhle Manzini and Lindokhule Manzini.
Education
Sipho was introduced to acting by a London teacher while he was still at Star of the Sea High School in uMhlabuyalingana. After finishing his matric he worked at a metal company in Johannesburg . He worked and managed to gather funds for his studies of Public Administration at Unisa. Manzini had a passion for acting and he then begin to take part in auditioning and later joined Sibikwa Arts Centre in Benoni to broaden his expertise. He is now a board member at this centre.

His first acting role was on Sokhulu and Partners on SABC1 AS Uncle Ntsiki on season 3. He made a remarkable debut in South African series such as Muvhango, Isibaya, Scandal and Rhythm City as Zachariah. He played a role as a celebrity guest in the second season of a film series titled Karektas.
Another noticeable role of Sipho was the role of he portrayed on Generations as Diamond the corrupt cop. On Ring of Lies he played the character of Sifiso. He played a character of Mr Zondo on the film Broken Vows, season 2. He also debuted as Bra Steve in season 5 of eKasi: Our Stories on season 2 of a situation comedy film titled Family Bond as a guest star. Again, he played a role of Governor Makhafula in the film called Lockdown.
